PAX Stage-tip


Usually, development of a new biotherapeutic entails developing a new method for analyzing various properties, such as glycosylation.  Glycans are instrumental in cellular signaling, therefore a specific protein's glycosylation can influence the effectiveness of the biotherapeutic.  Given the importance of glycosylation to overall performance of a glycoprotein, there is a need to completely characterize and then monitor these glycans for both research and manufacturing purposes. Many recombinant proteins, such as recombinant human erythropoietin (rhEPO), have highly intricate glycosylation patterns consisting of O-linked glycosylation as well as complex, branched N-linked glycosylation with three different N-linked glycosylation sites. The heterogeneity of N-linked glycosylation is mainly attributed to differences in sialylated glycans, which make approaches such as intact mass by high resolution mass spectrometry virtually impossible. This presents a challenge as the presence for sialic acid is critical in the characterization of a sialylated glycoprotein, as well as to removal of excess labeling reagent, salts, or surfactants.


The Preciex® PAX enrichment Stage-tip uses an anion exchange Glycocapture Resin to capture high-mannose, hybrid, and complex glycans from cleavage intact protein for effective downstream analysis by FLR, MALDI and ESI-MS. Traditional sample preparation involves intensive manipulation and a loss of sample, but with a PAX stage-tip product, the results are sensitive, effective, and save time. The benefit of strong hydrophilic interaction is critical to bind fluorophore-labeled glycans, like high polar sialylated glycans. 

Process of use: centrifugation, vacuum, positive pressure.
